    Phenotype

    Homozygous: There is no phenotype related to the homozygous floxed mouse without Cre. When Cre is present, the mutation causes accumulation of polyunsaturated fatty acids in circulating LDL, whereas wild-type mice will predominately have monounsaturated fatty acids in circulating LDL.

    Hemizygous: When Cre is present, the heterozygotes have half the ACAT2 activity of a Cre negative mouse.

    Strain Development
    The Soat2-targeting vector was electroporated into mouse-derived embryonic stem cells (likely of 129S5/SvEvBrd or 129S6/SvEvTac origin). The selected clones were injected into C57BL/6N blastocysts. Acat2-floxed mice were crossed with B6.129S4-Gt(ROSA)26Sortm1(FLP1)Dym/RainJ (RRID:IMSR_JAX:009086), resulting in the excision of the neo cassette and creating Acat2 floxed mice. The homozygous floxed mouse was then crossed with the Alb-Cre mouse, B6.Cg-Speer6-ps1Tg(Alb-cre)21Mgn/J (RRID:IMSR_JAX:003574), bred back to homozygosity for the floxed allele and maintained with sib-matings in which only one sibling was Cre-positive.

    1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

    2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients.
