Type: Gene
Species: Mouse
Chromosome: 11
Alteration at locus: Knockout

Genetic Alterations
The part of exon 2 that encodes the homeobox and the exon 2 splice acceptor site was replaced with a neo cassette.
ES Cell Line
E14 derived from 129P2/OlaHsd
Phenotype

Homozygous: Mice lack the first pair of ribs, mice exhibit an anterior transformation with vertebra C6 through T3 exhibiting the characteristics of C5 through T2, cultured bone marrow cells form more erythroid colonies consisting of erythroid burst-forming unites (BFU-E) and erythroid colony forming units (CFU-E) than wild-type bone marrow cells, and fetal liver cells generate more erythroid burst-forming units than cells from wild-type mice. Also can have eye problems (more pronounced with more 129 genetic background) and self-scratching behavior with age.

Heterozygous: Minor skeletal defects in a small percentage of heterozygotes. This mutation has no observable phenotype on C57BL/6J background.

Strain Development
Correctly targeted E14 ES cells derived from 129P2/OlaHsd were injected into blastocysts to generate chimeras. Chimeras were crossed to C57BL/6J and the resulting offspring were continuously bred back to C57BL/6J for 8 generations. Mice were then mated to generate a homozygous allele for the Hoxb6 mutation, and then later crossed to 129S6/SvEvTac for 4 generations. Subsequently they have been maintained by sib-mating. The MMRRC also maintains this allele on a C57Bl/6J background, MMRRC:037402, B6.129P2-Hoxb6tm1Cka/Mmucd.
Suggested Control Mice
Wild-type littermates or C57BL/6J mice
MMRRC Genetic QC Summary
The MMRRC Centers have developed a genetic QC pipeline using MiniMUGA array genotyping to provide additional information on strain backgrounds for MMRRC congenic and inbred strains. For more information on when data may be available, or to request genotyping for a strain of interest, please contact mmrrc@ucdavis.edu. Older strains may not have this information.

Colony and Husbandry Information

Colony Surveillance Program and Current Health Reports

Mice recovered from a cryo-archive will have health surveillance performed on recipient females. Health reports will be provided prior to shipment. If you require additional health status information, please email mmrrc@ucdavis.edu.
Coat Color
Black
MMRRC Breeding System
Backcross or Sib-mating
Breeding Scheme(s)
  • Backcross
    • Wild-type (C57BL/6J) female x Heterozygous male or reciprocal mating
  • Sib-mating

    • Homozygous female x Homozygous male littermate
Overall Breeding Performance
Good. Homozygous mutant Hoxb6 null females are not good mothers; they are fertile, but the pups would have to be cross-fostered.

Order Request Information

Limited quantities of breeder mice (recovered litter) are available from a cryoarchive; recovered litter usually available to ship in 3 to 4 months.

Cryopreserved material may be available upon request, please inquire to mmrrc@ucdavis.edu for more information.

Distribution of this strain requires submission of the MMRRC Conditions of Use (COU). A link to the COU web form will be provided via email after an order has been placed; the form should be completed then or the email forwarded to your institutional official for completion.

The donor or their institution limits the distribution to non-profit institutions only.

Additional charges may apply for any special requests. Shipping costs are in addition to the basic distribution/resuscitation fees. Information on shipping costs and any additional charges will be provided by the supplying MMRRC facility.

MMRRC Item # Description Distribution Fee / Unit (US $)
*Shipping & Handling not included*
Units Notes
037471-UCD-EMBRYO Cryo-preserved embryos $1,038.00 / Non-Profit Aliquot Approximate quantity2 : 20-40 embryos / aliquot
037471-UCD-SPERM Cryo-preserved spermatozoa $546.25 / Non-Profit Aliquot Approximate quantity3
037471-UCD-RESUS Litter recovered from cryo-archive $4,044.00 / Non-Profit Litter Recovered litter4; additional fees for any special requests.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.

4 The distribution fee covers the expense of resuscitating mice from the cryo-archive; you will receive the resulting litter.
