Genetic Alterations
A targeting vector was designed to flank exons 4 and 5 with loxP sites. Deletion of these exons would cause a frame shift, resulting in a stop codon after eight missense amino acids in exon 6.

Phenotype

Homozygous: No observable phenotype

Cre-excised Phenotype: Exposure of NOD.B10 COX-2flox splenocytes to recombinant HTN-Cre results in dose-dependent deletion of the floxed Ptgs2 gene within one hour of exposure. Earlier work showed that targeted in vivo expression of Cre in myeloid cells of C57BL/6 mice can block Ptgs2 expression in macrophages and attenuate acute inflammation. One objective of this laboratory is to discern whether B cell-targeted Cre excision of COX-2flox affects B cell function in vitro and B cell-driven autoimmune disease in vivo. Unfortunately, recent efforts to backcross a CD19cre gene onto the NOD.B10 background resulted congenic mice which failed to manifest Sjogren's syndrome hyposalivation (note: this occurred in the absence of COX-2flox); additionally, the latter strain failed to breed well and was lost. Further efforts in this direction would require backcrossing another gene for B cell-targeted Cre expression.

Strain Development
A floxed Ptgs2 gene LW1 derived from 129S4/SvJae was developed by and bred onto a 129xB6 background. The germline floxed Ptgs2 allele was maintained on a mixed 129xB6 background prior to being backcrossed onto the Sjogren's syndrome-susceptible NOD.B10Sn-H2b/J background through speed-congenic breeding. Because the Ptgs2 gene was localed in the center of an earlier mapped Aec2 disease susceptiblity region for Sjogren's syndrome in NOD.B10Sn-H2b/J mice, additional backcross generations were needed to recover the SS xerostomia phenotype.

Colony and Husbandry Information

This mouse strain appears to have a nervous disposition. This might explain why breeding has not been as routinely successful at the Feinstein Institute as at Charles River breeding facility (categorized as good). It might also explain why we have seen evidence of cannibalism of the litters here at the Feinstein Institute. The individual in charge of their breeding at Charles River GEMS (Karen.Dorman@crl.com) has indicated that some (but not all) of the breeders declined in productivity after 1-2 litters. Because of this, she breeders were typically replaced after 3-4 months of breeding.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
