Genetic Alterations
This is a CRISPR mediated knock-in, replacing endogenous Ascl1 (Gene ID:17172) with the same gene that has been altered to add two TdTomato (GenBank:AY678269) open reading frames separated by self-cleaving peptide 2A sequences (i.e., Ascl1-2A-TdTomato-2A-TdTomato), enabling expression of individual proteins, TdTomato and Ascl1.

Homozygous: TdTomato fluorescence in Ascl1 expressing cells. No effect on viability or fertility.

Hetero/Hemizygous: TdTomato fluorescence in Ascl1 expressing cells. No effect on viability or fertility.

Strain Development
Superovulated FVB females were harvested for embryos, and Cas9 mRNA, gRNA, and linear donor plasmid were microinjected. Embryos were transplanted to Swiss Webster foster females. Line has been maintained and backcrossed on the FVB background for 2 generations. The line is kept as heterozygotes, although for no particular reason.
