Genetic Alterations
A bacterial artificial chromosome (BAC; address RP23-117B6) was modified to hold a cre recombinase transgene (NCBI Gene ID:2777477) under the control of the Zic4 promoter (MGI:107201). The cre recombinase possessed a poly-A tail to prevent expression of the Zic4 gene from the BAC.

Phenotype

Homozygous: The submitter does not recommend that the Zic4-Cre (NIDA016) mice be bred to homozygosity

Hemizygous: If a Zic4-Cre (NIDA016) mouse is crossed with a mouse possessing a floxed allele, recombination is expected to occur in mitral neurons in the main and accessory olfactory bulb, scattered neurons in the striatum, granule neurons in the cerebellum, and neurons in the thalamus and septum. Bergmann glia in the cerebellum will also display recombination, as will some scattered glia in various brain regions.

Strain Development
The RP23-117B6 bacterial artificial chromosome was modified as described in PMID:20885380. The background of the embryos that were injected was C57BL/6J (JAX Strain #00664). All subsequent generations were maintained on this background.

The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
