Genetic Alterations
Deletion of exon 1 in the chromatin remodeling gene Cecr2 (MGI:1923799) via floxing & homologous recombination. The exon was removed by cre-mediated recombination (BALB/c-Tg(CMV-cre)1Cgn/J - Jackson Strain #003465). Originally designed to be conditional, however, that part of the design was unsuccessful.

It can be used in combination with our hypomorphic genetrap mutation, Cecr2Gt(pGT1)1Hemc (MMRRC:43819 and MMRRC:43821).

    Phenotype

    The phenotype arising from this mutation is extremely background-dependent (e.g., low penetrance on FVB background - see MMRRC:43820). On BALB/c, penetrance is complete and the phenotypes are:

    • Heterozygotes: Exencephaly (MP:0000914) at a very low penetrance (~1-3%). Kinked tail at higher penetrance (MP:0000585).
    • Homozygotes: Exencephaly (MP:0000914), kinked tail (MP:0000585), lethality.
    Loss of Cecr2 causes exencephaly, reduced fertility in both males and females, and kidney defects (duplex kidney), all in a strain-dependent manner. For this deletion mutation, exencephaly has a penetrance of ~100% on the BALB/cCrl background and ~15% on the FVB/NJ background. The adult fertility defects can't be studied on the BALB/cCrlAlt background since there are no viable mutant adults, but the exencephalic embryos live to birth.

    Strain Development
    After crossing to BALB/c-Tg(CMV-cre)1Cgn/J - Jackson Strain #003465 to produce the deletion, the mutation was bred to N10 on BALB/cCrlAlt (heterozygous males bred with BALB/cCrlAlt females).
    MMRRC Genetic QC Summary
    Genetic QC analysis (GQC) was performed on representative samples from the donor submission and provides a baseline reference. Regardless of the material ordered (live mice, sperm, embryos, resuscitated animals), the provided material is guaranteed to contain the primary genetic alteration of interest; however, the composition of the genetic background may vary (See Reproducibility sections of the report for details). The impact of genetic background on previously reported phenotypes has not been evaluated.

    GQC analysis was done on two samples AE1055 (XX), CY7776 (XY) at the time of importation and the conclusions are consistent in all samples. The results of the analysis are fully consistent with the SDS and CY7776 is the representative sample shown in the report. The primary (host) background is consistent with the BALB/c strain group. A BALB/cCrlAlt substrain reference is not available in miniMUGA and BALB/cAnNCrl is the closest reference strain. The secondary (donor) background is consistent with C57BL/6NTac, which is in agreement with the background of the ES cell line used- iTL BA1 (C57BL/6NTac x 129S6/SvEvTac)F1). The mitochondrial genome and the Y chromosome are consistent with both the host and donor background (identical by descent). The strain is 99.75 % inbred and 100 % congenic. The region containing the allele of interest has the expected donor background (C57BL/6NTac). None of the constructs detectable by MiniMUGA are present.

    The genome of the MMRRC: 43818 strain is 99.75 % inbred and 100% congenic at the strain level. 0.25% of the genome is segregating for the secondary (donor) background. The level of congenicity estimated by miniMUGA is consistent with the number of backcross generations reported in the SDS (10 Generations).

    Colony and Husbandry Information

    Heterozygous crosses are possible, but the pure BALB/cCrlAlt appear to be better breeders. The strain's donor adopted the approach of crossing het x het when mutant embryos were needed. For stock, the donor crossed het males to pure BALB/cCrlAlt females. The difference is small and het x het crosses would be fine, however, mutant pups are lost at birth. 

    Generally, the litters are average/small (~6 pups); parenting is excellent. Breeding in trios is recommended to regularly get all pups in both litters to wean.

The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary.
