Genetic Alterations
The gene trap vector including the En2 intron and Beta-geo-SV40pA fell within intron 7 or 8 of the locus, depending on alternative splicing of exon 8. It was mapped to the middle of intron 7. Mutated CT45 ES cells containing this vector were used to generate mutant mice.

    Phenotype
    Homozygous - Exencephaly (MP:0000914), kinked tail (MP:0000585), lethality. Mutant animals which are non-penetrant for exencephaly show reduced fertility (males - MP:0001922 - and females - MP:0001923) and kidney defects (MP:0004017).

    Heterozygous/Hemizygous - Exencephaly (MP:0000914) at a very low penetrance (~1-3%). Kinked tail at higher penetrance (MP:0000585).

    Loss of CECR2 causes exencephaly, reduced fertility in both males and females, and kidney defects (duplex kidney), all in a strain-dependent manner. For this hypomorphic mutation, exencephaly has a penetrance of 50% on BALB/cCrl background. Fertility defects appear more prominent on BALB/cCrl background.

    Strain Development
    The mutation was bred to N10 on BALB/cCrlAlt (see MMRRC:43818). It was also bred onto C57BL/6 and 129P2, but those lines no longer exist. Until submission, the strain was bred with BALB/cCrlAlt crossed with a heterozygote or by crossing two heterozygotes.

    1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

    2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

    3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
