The strain was closed to outside B6 strains in the early 2000s and has been maintained as a closed colony. It carries approx. 300 private homozygous variants that distinguish it from the original JAX line (JAX Strain #002020).

Homozygous: lethal

Hetero/Hemizygous: intestinal neoplasia, adenomas develop primarily in the small intestine with the preponderance of the adenomas developing in the distal half. MMRRC:43849 and MMRRC:50543 are genetically identical but harbor two distinct complex microbiota that alter the intestinal adenoma phenotype. The original ApcMin mice from the closed genetic colony from William Dove at the University of Wisconsin was rederived using surrogate dams that harbored two distinct microbiota populations: low complexity and high complexity. The original closed colony line is available through the MMRRC with either the low complexity (MMRRC:43849) or high complexity (MMRRC:50543) microbiota that have the represented phenotypes in the small intestine and colon.

Colony and Husbandry Information

The original ApcMin mice from the closed genetic colony from William Dove at the University of Wisconsin was rederived using surrogate dams that harbored two distinct microbiota populations: low complexity and high complexity. The original closed colony line is available through the MMRRC with either the low complexity (MMRRC:43849) or high complexity (MMRRC:50543) microbiota that have the represented phenotypes in the small intestine and colon. Crossing, co-housing, or mating to other mouse lines will potentially alter the microbiota and phenotype of this line. Littermate controls should be used to ensure appropriate scientific rigorous results.
Animals are maintained on breeder chow (5058 PicoLab Mouse Diet 20).

Rigor and Reproducibility Considerations (e.g., background strain genetics, microbiota characterization and influence)

Statistical testing demonstrates that both MMRRC:43849 and MMRRC:50543 develop significantly more tumors than ApcMinmice from the Jackson Laboratory (Strain #002020). MMRRC:50543 (high complexity microbiota) develop significantly more tumors than MMRRC:43849 (low complexity microbiota). While MMRRC:50543 has similar tumor numbers to ApcMinmice from McCardle Laboratory for Cancer Research, MMRRC:43849 has significantly fewer tumors than ApcMin from McCardle.

MMRRC Breeding System
Random intra-strain mating within a specific microbiota profile. Complex gut microbiotas have been maintained at our facility by housing mice in ventilated racks, occasionally mixing bedding among colony animals and changing gloves prior to working with animals in this colony. Periodic (e.g., annual) monitoring of gut microbiota is also recommended as institutional drift may occur with time for, as yet, undetermined reasons.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. 2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
