Genetic Alterations
An FRT-flanked neo cassette and loxP site were inserted upstream of Ugdh exon 6 and a loxP site was inserted downstream of exon 6 via homologous recombination. Flp-mediated recombination removed the neo cassette.

Strain Development
The targeting vector was constructed using the recombineering method from a 12.7-kb genomic fragment, which was retrieved from a C57BL/6 BAC clone (RP23-477N9, BACPAC Resources Center at Children's Hospital Oakland Research Institute). It contains a neomycin (Neo) selection cassette surrounded by two FRT sites and exon 6 of the Ugdh gene flanked by two loxP sites. The linearized targeting construct was electroporated into an ES cell line of 129S6/SvEvTac origin. Recombinant clones were screened by Southern blot analysis with 5’ (SacI) and 3’ (BamHI) external probes before being injected into C57BL/6 blastocysts. Chimeras were further bred with C57BL/6 mice for germline transmission, which was confirmed by PCR genotyping using the following primers: UgdhfloxF: 5’-TTCTGAGGCTGTATTTCACTTCC-3’; UgdhfloxR: 5’-AGGCACAGGCACGATTAGGA-3’. Amplification bands of 214 bp and 314 bp correspond to the wild-type and floxed alleles, respectively. The FRT-flanked Neo cassette was removed by crossing with an FLP-transgenic line (B6.129S4-Gt(ROSA)26Sortm1(FLP1)Dym/RainJ - JAX Strain #009086).

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
