Phenotype
In these Notch1-deficient mice, somitogenesis is disturbed and there is a probable defect in the chorio-allantoic connection. Homozygotes die at embryonic day 9. By 2 months of age, mild aortic root dilation was observed in a subset of the heterozygotes in comparison with wild-type littermates. Time course studies through 9 months of age revealed 100% of het mice displayed aortic root dilation in comparison with wild-type littermates. Histologic examination of the het aorta at 9 months of age revealed localized thinning of the aortic wall. Within the thinned region, smooth muscle cells had abnormal morphology with dysmorphic, rounded nuclei and a reduction in elastic laminae, demonstrating that heterozygous deletion of Notch1 causes ascending aortic aneurysms in mice. Of note - aortic root dilation was not observed in hets in a mixed background and het mice did not have a bicuspid aortic valve.

Strain Development
Mice heterozygous for Notch1 allele were purchased from Jackson Laboratories (JAX Strain #002445). These mice were initially backcrossed into C57BL/6J background for >10 generations. Subsequently, the donor backcrossed these mice into 129S6/SvEvTac for 6 generations.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
