    Phenotype
    Rgs21-null mice have blunted aversion to quinine and denatonium, and blunted preference for monosodium glutamate, the sweeteners sucrose and SC45647, and (surprisingly) NaCl.

    Taste perception phenotypes are only elaborated when Cre-recombinase is cross-bred and expressed (or induced).
    Strain Development
    Conditional knockout of the Rgs21 gene was produced directly in the C57BL/6 background by genOway (Lyon, France). It involved insertion of loxP sites on either side of Rgs21 exon 5 (which encodes the final 67 amino-acids of the 152-amino acid RGS21 open-reading frame), as well as a neomycin-resistance gene (neo) flanked by “flippase recognition target” (FRT) sites, by homologous recombination in C57BL/6 embryonic stem cells. Drug-selected embryonic stem cells, with PCR and Southern blot evidence of homologous recombination, were subsequently injected into C57BL/6 blastocysts. Resulting chimeric male mice were crossed with females from the genOway proprietary ubiquitous Flp recombinase expressing mice for in vivo removal of the FRT-flanked neo selection cassette. F1 heterozygous Rgs21fl/+ mice devoid of the neo selection cassette were delivered by genOway to the Siderovski lab at WVU. This conditional ‘floxed’ Rgs21 knockout strain was maintained at WVU by backcrossing to the C57BL/6J strain.

    MMRRC Genetic QC Summary
    Genetic QC analysis (GQC) was performed on representative samples from the donor submission and provides a baseline reference. Regardless of the material ordered (live mice, sperm, embryos, resuscitated animals), the provided material is guaranteed to contain the primary genetic alteration of interest; however, the composition of the genetic background may vary (See Reproducibility sections of the report for details). The impact of genetic background on previously reported phenotypes has not been evaluated.

    GQC analysis was done on two XY samples, WA9288 and JK5682, at the time of importation and the conclusions are consistent in all samples. The results of the analysis are congruent with the SDS and WA9288 is the representative sample selected for the report. The primary (host) background is C57BL/6 and there is positive evidence of the C57BL/6J and C57BL/6N substrains. The mitochondrial genome is consistent with the C57BL/6 background and the Y chromosome is consistent with at least one backcross to C57BL/6J. Both samples are homozygous for the C57BL/6N background at the locus. The following constructs are detected in all samples: IRES, r_FP?.

    The genome of MMRRC:046280 mice is 87% inbred at the substrain level. 13% of the genome is segregating for the two substrains,C57BL/6J and C57BL/6N. The contribution of C57BL/6J is 86.3%. The contribution of C57BL/6N is 13.7%.

    The estimated inbreeding, congenicity, and the relative strain/substrain contribution depend on the number of samples genotyped. The genomes of strains that are not fully inbred are not completely reproducible.

    ATTENTION: MiniMUGA has detected a potential unexpected construct (r_FP) that may interfere with the experimental design/rigor and reproducibility. It is recommended to genotype to ensure that r_FP is not present.

    Based on this report, a request has been made to update the strain name from C57BL/6J-Rgs21tm1.1Dski/Mmnc to C57BL/6-Rgs21tm1.1Dski/Mmnc
