Genetic Alterations
The 280-kb deletion of the region in between Ins2 and Ascl2, and including Th, was induced in vivo by Cre-recombination using TAMERE.

Phenotype
Maternal transmission leads to a hypomorphic Ascl2 phenotype characterized by intrauterine growth restriction and abnormal placentation. The parietal giant cell layer is expanded and the junctional zone is severely hypoplastic. As a consequence, the labyrinth layer is thickened and expands proximally up to the giant cell layer. Upon paternal transmission, Del7AI leads to defects in the silencing of the paternal allele of Ascl2 and Tssc4. This loss of imprinting model is able to rescue maternally inherited Ascl2 mutations. Again, Ascl2 levels are very low, growth is affected and the placentas are disorganized. This led us to characterize the role of ASCL2 in the glycogen trophoblast cell lineage.

Strain Development
Conditional and complementary PGK-neo-pA cassettes were independently targeted 3' of Ascl2 (MGI:4399141) and 5' of Ins2 (MGI:3805819) in R1 ESCs. The insertion at Ascl2 is PGK-loxP-neo-pA-loxP, the one at Ins2 is loxP-PGK-loxP-neo-pA. Mice trans-heterozygous for these alleles and carrying a Sycp1-Cre transgene (MGI:2176068, PMID:9771647) were bred and the 280-kb Del7AI deletion allele was obtained by trans-allelic recombination in spermatocytes using the TAMERE approach (PMID:9843213). The structure of the deletion breakpoint is Ascl2-PGK-loxP-neo-pA-Ins2. The mutation has been maintained on the outbred CD1 strain background.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
