Genetic Alterations:
The Tlr7 gene on
the X chromosome of rsq2 mice was sequenced, and an A to T transversion
was identified in exon 3 (of 3 total exons) at position 732. The mutation
results in a conversion of asparagine to tyrosine at residue 182 of the
TLR7 protein.

Phenotype:
Homozygous phenotype: Rsq2 is X-linked.
Heterozygous females are normal, but hemizygous males have abolished TLR7
signaling
Hemizygous phenotype: The
rsq2 mutant was discovered in a screen of ENU-mutagenized G3 mice for
altered responses to Toll-like receptor (TLR) ligands (TLR Signaling
Screen). Peritoneal macrophages from rsq2 mice fail to produce tumor
necrosis factor (TNF)-? in response to R-848, a single stranded RNA
mimetic that activates TLR7. However, TNF-? production is normal
following stimulation with MALP-2 (macrophage-activating lipopeptide-2,
TLR2/6 ligand), CpG oligodeoxynucleotides (CpG ODN, TLR9 ligand),
Pam3CSK4 (a triacyl lipopeptide, TLR2/1 ligand), poly I:C (TLR3 ligand)
and lipopolysaccharide (LPS, TLR4 ligand).

TLR7 signaling is abolished. The mice will probably be sensitive to certain infections but are healthy under normal conditions.
Mice recovered from a cryo-archive will have health surveillance performed on the
resuscitated animals.
Limited quantities of breeder mice (recovered litter) are available from a cryoarchive; recovered litter usually available to ship in 3 to 4 months.

a The distribution fee covers the expense of resuscitating mice from the cryo-archive; you will receive the resulting litter. The litter will contain at minimum one breedable mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC repository will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC repository.
b An aliquot contains sufficient material (volume and concentration) for at least two IVF or several artificial inseminations (based on our procedures). The MMRRC makes no guarantee concerning success of these procedures when performed outside the MMRRC facilities.
c An aliquot contains a sufficient number of embryos (in one or more vials and based on the transfer success rate of the MMRRC facility) to transfer to at least two recipients.
