Genetic Alterations:
The long arm of homology in the targeting vector was a 4.5 kb NotI/EcoRI fragment upstream of the IS4 region of alpha1 3.2 subunit, which has been subcloned into pBluescript KS (+) (pBS) and cut with NotI/ClaI. The short arm was a 2.2 kb BamHI/BamHI fragment downstream of the extracellular loop between IS5 and IS6. These fragments were inserted into cloning sites of pPNT flanking a reversed PGK-neomycin resistance cassette. The vector included a thymidine kinase cassette distal to the short arm to allow for negative selection of non-recombinants.

Strain Description

Phenotype:
Homozygous phenotype: alpha1 3.2-null mice have constitutively constricted coronary arterioles and focal myocardial fibrosis. Coronary arteries isolated from alpha1 3.2-null arteries showed normal contractile responses but reduced relaxation in response to acetylcholine and nitroprusside.

Heterozygous phenotype: Heterozygotes do not show any morphological abnormalities.

Special Considerations

The use of mouse houses help to facilitate better reproductive results. Once the pups are weaned at 21 days, they should be grueled for about one week.

Control Mice:

  • Wildtype littermates

1 The distribution fee covers the expense of resuscitating mice from the cryo-archive; you will receive the resulting litter. The litter will contain at minimum one breedable mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains sufficient material (volume and concentration) for at least two IVF or several artificial inseminations (based on our procedures). The MMRRC makes no guarantee concerning success of these procedures when performed outside the MMRRC facilities.

3 An aliquot contains a sufficient number of embryos (in one or more vials and based on the transfer success rate of the MMRRC facility) to transfer to at least two recipients.
