Genetic Alterations
Thyrotropin receptor (TSHR) A-subunit cDNA (amino acids 1-289) with a C-terminal 6 histidine tag was excised from pSV2-ECE-TSHR-289-6H-dhfr with EcoR I and Xba I and subcloned into pcDNA3. This intermediate plasmid (pcDNA3- 289), containing a Kpn I site 5' to the insert, was mutagenized to introduce a Kpn I site 3' to the bovine growth hormone polyA tail. The bovine thyroglobulin (Tg) promoter in the plasmid TgSK+ (amino acids -2036 to +9; Gene Bank accession No. M35823) was restricted with BamH I; the released 2 kb fragment was inserted in the correct orientation into pcDNA3-289 to generate pTg-TSHR-289. After excision with Kpn I, purified pTg-TSHR-289 was micro-injected into CB6F1 x CB6F1 oocytes.

Phenotype
Homozygous: Not studied

Hemizygous: Normal thyroid architecture and serum thyyroxine (Tg). Unlike wild-type littermates of control BALB/c, unresponsive to low dose immunization with A-subunit adenovirus and responds (albeit at low level) to a high dose of A-subunit adenovirus. Not tested by conventional immunization. When assayed for intrathyroidal protein, line 51.9 has the lowest human A- subunit protein protein content (highest in line 50.6, followed by 60.6).

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
