Genetic Alterations
The moonlight mutation was mapped to Chromosome 4. Sequencing of the critical region found a deletion spanning exons 22-28 (of 48 total exons) of the Dock7 gene (positions 118824 to137848 occurring in the middle of introns 22 and 29 in the Genbank genomic region NC_000070 for linear genomic DNA sequence of Dock7). If the donor and acceptor splice sites of introns 22 and 29 are still used, amino acids 866-1173 of the DOCK7 protein would be deleted, and a frameshift and insertion of 30 aberrant amino acids before a premature stop codon would occur.
Phenotype
Homozygous: The moonlight mutation was induced by ENU mutagenesis on the C57BL/6J background, and was discovered in G3 animals. Moonlight mice have a slightly lightened coat, white-tipped paws, and hypopigmented genitalia. Moonlight animals display an occasional belly spot similar to splotch2 mutants, and sometimes a white-tipped tail. Moonlight females are poor mothers, and their pups require fostering.

Strain Development
The moonlight mutation was induced by ENU mutagenesis on the C57BL/6J background, and was discovered in G3 animals. All subsequent crosses to maintain strain were on a C57BL/6J background.

Colony and Husbandry Information

Homozygous females have problems nurturing young; pups require fostering. Because of poor homozygous female breeding, it is best to breed homozygous males with heterozygous females.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.

4 The distribution fee covers the expense of resuscitating mice from the cryo-archive; you will receive the resulting litter.
