Genotype Determination
Phenotype
Hemizygous DCX-CreERT2 mice are viable and fertile, with CreERT2 fusion gene expression directed by the human doublecortin (DCX) promoter/enhancer regions. The donating investigator reports that no abnormal phenotype is observed in hemizygous mice. Cre-ERT2 fusion gene activity is inducible; observed following taxomifen administration. As such, when DCX-CreERT2 mice are bred with mice containing loxP-flanked sequences, tamoxifen-inducible Cre-mediated recombination will result in deletion of the floxed sequences in the DCX-expressing cells of the offspring. The donating investigator reports tamoxifen-inducible Cre recombinase activity is observed throughout the central nervous system, including most brain regions and spinal cord. The donating investigator did not examine Cre-ERT2 activity in tissues other than brain, or in any tissues prior to tamoxifen treatment.
Strain Development

A transgene was designed with the human doublecortin (DCX) promoter/enhancer regions upstream of a CreERT2 fusion gene (Cre-ERT2; Cre recombinase fused to a G400V/M543A/L544A triple mutation of the human estrogen receptor ligand binding domain). This transgene was microinjected into C57BL/6J blastocysts. Transgenic mice from a single founder line were bred to C57BL/6J mice. These DCX-CreERT2 transgenic mice were subsequently maintained by breeding hemizygous mice with C57BL/6J inbred mice for many generations prior to arrival at the MMRRC.

A sample from this MMRRC strain was analyzed using the Mouse Universal Genotyping Array (MUGA) and MMRRC computational tools were used to assess the genetic background. A summary of the data can be found here.
