Genetic Alterations
The wild-type full-length human PC-1 q allele was subcloned into plasmid pRK7 under the control of the CMV promoter, as previously described (11). Transgenic animals were made as follows. Gene constructs were freed of plasmid sequences by gel electrophoresis after appropriate restriction enzyme digestion. The DNA was injected into the pronucleus of fertilized mouse eggs harvested from recently mated and superovulated, 4- to 6-wk-old female, white FVN mice and black B6D2 mice. After the animals were euthanized, the recovered eggs were allowed to recover, and the DNA was then injected into pronuclei. Phenotype
Homozygous: Homozygous PC-1+/+ mice have improved glucose tolerance and insulin sensitivity compared to wild type.

Heterozygous/Hemizygous: Heterozygous PC-1+/+ mice have improved glucose tolerance and insulin sensitivity compared to WT, but diminished glucose tolerance and insulin sensitivity compared to homozygous PC-1+/+.
Strain Development
Originally the human PC-1 transgene to Q allele was introduced into FVB/N stain because it is easier to obtain transgenic mice from this strain. We obtained 6 founders, with 1 line having high PC-1 expression in both liver and muscle. The F1 generation was bred with C57B6 to produce a F1 hybrid (F1H). The PC-1 transgenic hybrids were bred with C57B6 until F3H generation and then were inbred for 4 more generations.

Colony and Husbandry Information

Tg male or female backcrossed to B6 male or female. All breeding pairs and their pre-wean litters are housed in shoebox cages with water bottles and receive cotton nestlet. They receive a standard normal lab rodent chow. All non-breeding mice are housed in shoebox cages with water bottles and nestlet; they also receive a standard normal lab rodent chow.

Colony Surveillance Program and Current Health Reports

Mice recovered from a cryo-archive will have health surveillance performed on recipient females. Health reports will be provided prior to shipment. If you require additional health status information, please email mmrrc@ucdavis.edu.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
