Genetic Alterations
The Ple155-lacZ transgene (pEMS1626) was designed with the 1652 bp Ple155 MiniPromoter (derived from a portion of the upstream promoter of the PCP2 gene (prior to exon 1) and the downstream second promoter of the PCP2 gene (prior to exon 2) from the Purkinje cell protein 2 (PCP2) gene) upstream of a frt-flanked β-galactosidase (lacZ) gene, an SV40 early polyA signal, and a human HPRT complementary sequence (containing exon1, intron1, exon2, and part of intron2). This construct was targeted as a single copy knockin to the Hprtb-m3 mutant locus on the X chromosome via electroporation into mEMS1202 embryonic stem cells (generated from F1 generation male pups [heterozygous for the Hprtb-m3 allele and wild-type at the Gt(ROSA)26Sor locus] born from C57BL/6-congenic females homozygous for a Hprtb-m3 mutation [originally derived from 129P2/OlaHsd-derived E14TG2a embryonic stem cells] and 129S-Gt(ROSA)26Sortm1Sor heterozygous males.
ES Cell Line
mEMS1202 derived from (B6.129P2-Hprtb-m3/J x 129S-Gt(ROSA)26Sortm1Sor/J)F1
Phenotype

Homozygous: Not evaluated

Heterozygous: These Ple155-lacZ;mESM4473 mice have the Ple155-lacZ transgene targeted as a single copy "knockin" into the upstream region of hypoxanthine guanine phosphoribosyl transferase (Hprt) locus on the X chromosome. Heterozygous females and hemizygous males are viable and fertile, with the promoter/regulatory regions of the Purkinje cell protein 2 (PCP2) gene directing expression of β-galactosidase (lacZ). These Ple155-lacZ;mESM4473 mice may be useful in studying PCP2-expressing cells in the brain and diseases affecting the brain.

Of note, Ple155-EGFP;mEMS1738 mice have expression of EGFP directed by the same MiniPromoter. The Ple155-EGFP;mEMS1738 mice are described by the donating investigator as: No EGFP expression was detected above background in native fluorescence under epi or confocal microscopy. Anti GFP immunostaining (rabbit polyclonal, Invitrogen) did not detect specific GFP expression above background across numerous brain regions (brightfield).

Strain Development
Correctly targeted embryonic stem cells were microinjected into recipient mice cells. The resulting chimeric males (founder line mESM4473) were bred to B6(Cg)-Tyrc-2J/J females (B6-albino) to establish the mutant colony. The donating investigator reports that these Ple155-lacZ;mESM4473 mutant mice were bred with C57BL/6J wild-type mice for four generations prior to sending to the MMRRC. Upon arrival, Ple155-lacZ;mESM4473 mutant males were used to cryopreserve sperm. When rederiving a live colony, sperm will be used along with oocytes from C57BL/6J inbred mice to generate obligate heterozygous females, and these females will be bred at least one generation to C57BL/6J inbred mice to establish the colony.
Suggested Control Mice
Wild-type littermates or C57BL/6J mice
MMRRC Genetic QC Summary
The MMRRC Centers have developed a genetic QC pipeline using MiniMUGA array genotyping to provide additional information on strain backgrounds for MMRRC congenic and inbred strains. For more information on when data may be available, or to request genotyping for a strain of interest, please contact csmmrrc@jax.org. Older strains may not have this information.

Colony and Husbandry Information

The donating investigator recommends maintaining this strain by breeding heterozygous females with C57BL/6J inbred males.

Cryo-recovered strains distributed by the MMRRC at JAX are shipped to the customer from the Pathogen & Opportunistic-Free Animal Room G200 - see https://www.jax.org/jax-mice-and-services/customer-support/customer-service/animal-health/health-status-reports.

Mice recovered from a cryo-archive will have health surveillance performed on recipient females. Health reports will be provided prior to shipment. If you require additional health status information, please email csmmrrc@jax.org.
NOTE: "Hemizygote" as used here refers to males carrying a mutation on the X Chromosome or mice of either sex carrying an inserted transgene with no homologous allele on the other chromosome.

Order Request Information

Limited quantities of breeder mice (recovered litter) are available from a cryoarchive; recovered litter usually available to ship in 3 to 4 months.

Cryopreserved material may be available upon request, please inquire to csmmrrc@jax.org for more information.

Distribution of this strain requires submission of the MMRRC Conditions of Use (COU). A link to the COU web form will be provided via email after an order has been placed; the form should be completed then or the email forwarded to your institutional official for completion.

The donor or their institution limits the distribution to non-profit institutions only.

Additional charges may apply for any special requests. Shipping costs are in addition to the basic distribution/resuscitation fees. Information on shipping costs and any additional charges will be provided by the supplying MMRRC facility.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.

4 The distribution fee covers the expense of resuscitating mice from the cryo-archive; you will receive the resulting litter.
