Genetic Alterations
The transgenic construct contains the 201-kb P1 artificial chromosome (PAC) 61D6 and includes 5 kb of sequence upstream of the human MAPT promoter and an additional 62 kb downstream of the 3' end of MAPT. The downstream segment also contains the 3' end of the gene KIAA1267, however, human MAPT is the only complete gene in the PAC. The E10+14 mutation (PMID:10318930) results in the T-to-C exchange in intron 10 (NM_001123066.3:c.1920+14T>C). The mutation alters the alternative splicing of MAPT transcripts resulting in an increased 4R/3R tau ratio.

The endogenous, murine Mapt gene has been knocked out.

Phenotype
Mice that are homozygous for the targeted mutation and hemizygous for the transgene are viable, fertile, normal in size, and do not display any gross physical or behavioral abnormalities. These double-mutant mice produce mutated human tau (MAPT), but do not produce mouse tau (Mapt). The hT-PAC-E10+14 mouse model (with the E10+14 MAPT mutation) expresses all six human tau isoforms in the mouse brain with a [3-repeat (3R)-tau to 4-repeat (4R)-tau] isoform ratio of approximately 1:4, rather than the expected 1:1. The E10+14 mutation in humans increases exon 10 splicing and is associated with frontotemporal dementia with parkinsonism-linked to chromosome 17 (FTDP-17). No gross or microscopic pathologic lesions were observed in these mice. This mutant mouse strain may be useful in studies of tau isoforms in neurodegenerative disease. Heterozygous mice were not evaluated.

Strain Development
This transgene was microinjected into fertilized (C57BL/6 x C3H)F1 hybrid oocytes and founder mice were backcrossed to C57BL/6J mice for approximately 10+ generations. This transgenic line was crossed to B6.129X1-Mapttm1Hnd/J mice (JAX Strain #007251) to generate mice that were homozygous for the targeted mutation and hemizygous for the transgene. The colony has been maintained by sibling mating for three generations prior to donation to the MMRRC.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
