Genetic Alterations

This strain represents one of a set of 7 consomic strains in which progenitor inbred strains C57BL/6ByJ and 129P3/J were used to construct reciprocal chromosome substitution strains (CSSs) for Chr 1, 2, 7 and 9 using a "speed consomics" approach. In this strain, chromosome 7 was moved from donor strain 129P3/J to host strain C57BL/6ByJ.

Phenotype
The 129P3/J and C57BL/6ByJ progenitor inbred strains differ in taste responses, ingestive behavior, alcohol consumption, body size, adiposity, and other traits. Genome scans of crosses between these strains have detected QTLs for these phenotypes that cluster on Chr 1, 2, 7 and 9 (Adip2; Adip3, Adip5, Ap7q, Bwq5, Bwq6, Nattq1, Nattq2, Sucq).

Strain Development
Our breeding scheme was designed to eliminate Y chromosome from the donor strain. The 129P3/J and C57BL/6ByJ progenitor inbred strains were crossed and F1 females were backcrossed to males from a host strain, C57BL/6ByJ. Male offspring with a non-recombinant chromosome derived from the donor strain were identified in each backcross generation by genotyping and were backcrossed to females from the host strain, C57BL/6ByJ. To facilitate elimination of the donor, 129P3/J genome in non-target chromosomes using a "speed consomic" approach, we conducted genome scans of the N2 and N3 backcross generations (948 SNPs covering 19 autosomes and the X chromosome were genotyped to identify optimal breeders). Males with a non-recombinant donor chromosome and the smallest proportion of the donor genome in rest of the chromosomes were used as founders of the next generation. In subsequent backcrosses, donor chromosomes (1, 2, 7, 9) were genotyped with sets of SNP markers. We used the most centromeric and the most telomeric polymorphic markers available. To prevent loss of donor chromosome fragments due to double-crossovers, we calculated the cross-specific genetic recombination distances between markers and selected internal markers spaced no more than 10 cM. At the N6 backcross generation, males and females with the non-recombinant donor chromosome were intercrossed. Progeny of this intercross that are homozygous for the donor, 129P3/J chromosome was used to propagate the consomic strains.

The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. 2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. 3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
