Genetic Alterations
Insm1(GFP-Cre) mice contain a GFP/Cre fusion cassette that replaces the entire coding region of the Insm1 (insulinoma-associated 1) gene. Mice homozygous for the mutation are not viable. This strain may be useful for lineage tracing of Insm1+ cells and for studying pancreatic endocrine, neural and olfactory epithelium progenitor cells.

Phenotype
Insm1 (insulinoma-associated 1) encodes a protein containing both a zinc finger DNA-binding domain and a putative prohormone domain. It is expressed in pancreatic endocrine progenitor cells beginning at E9.5. Insm1 also is expressed in neural and olfactory epithelium precursor cells. Insm1(GFP-Cre) mice contain a GFP/Cre fusion cassette that replaces the entire coding region of the Insm1 gene. Mice homozygous for mutation are not viable. This strain may be useful for lineage tracing of Insm1+ cells and for studying pancreatic endocrine, neural and olfactory epithelium progenitor cells.

Strain Development
Using recombinase mediated cassette exchange (RMCE) a 532 bp fragment containing a GFP-Cre fusion protein replaced the entire coding region of the Insm1 gene. The donor construct includes a frt-flanked pgk-hygromycin immediately downstream of the GFP-Cre fusion sequence. The construct was electroporated into 129S6/SvEvTac derived embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts. Chimeric mice were bred with B6;SJL-Tg(ACTFLPe)9205Dym/J transgenic mice to delete the pgk-hygromycin cassette. The donating investigator reported that the mice were backcrossed to CD-1 mice for 14 generations prior to sending to MMRRC at JAX. Upon arrival at The Jackson Laboratory, mice were bred to C57BL/6J inbred mice for at least one generation to establish the colony.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
