Genetic Alterations
ENU-induced T to C transition at base pair 22,666,020 (v38) on chromosome 8, or base pair 40,566 in the GenBank genomic region NC_000074 for the Ikbkb gene, corresponding to residue 1,841 in the mRNA sequence NM_001159774 (variant 1) within exon 17 of 22 total exons and to residue 1,841 in the mRNA sequence NM_010546 (variant 2) within exon 17 of 21 total exons. The mutation results in an leucine (L) to proline (P) substitution at position 570 (L570P) in both isoforms of the inhibitor of kappa-B kinase-? (IKK-?; alternatively, IKK-2) protein.
Phenotype

Homozygous: Mice exhibited impaired IgE and IgG1 responses to the cysteine protease papain. Some mice also exhibited an increased viral titer in the liver after exposure to mouse cytomegalovirus (MCMV). Mice exhibited an increase in the CD4+ to CD8+ T cell ratio caused by a diminished frequency of peripheral CD8+ T cells in CD3 T cells after MCMV infection. Some mice also exhibited a reduced frequency of B1a cells in B1 cells and an increased frequency of B1b cells in B1 cells in the peripheral blood after MCMV infection.
