Genetic Alterations
This line carries a Rosa26 knock-in of an intersectional cassette consisting of a CAG promoter, an FRT-flanked neomycin resistance gene with STOP cassette, a LoxP-flanked STOP cassette, and a cDNA of H2B-TagBFP, sfGFP, and synaptophysin-tdTomato separated by P2A self-cleaving peptide cassettes.
ES Cell Line
AB2.2 derived from 129S7/SvEvBrd-Hprtb-m2
Phenotype
  • Homozygous: None expected.
  • Hetero/Hemizygous: None.
This mouse line is responsive to Cre and FLP recombinases. Upon Cre/FLP dual recombination, cells will express H2B-TagBFP (blue nuclei), sfGFP (green whole cell), and synaptophysin-tdTomato (red presynaptic contacts). It can be used to delineate cell types by dual expression of Cre and FLP recombinases through transgenic transmission or virus. Upon dual recombination, intersectional targeted cells will be marked with fluorescence in three areas of the cell: blue nuclei, green whole cell, and red presynaptic contacts. This is useful in studies on general neural morphology, circuit analysis, cell counts, and more.

Strain Development
  1. Generation of targeted AB2.2 ES cells through co-electroporation of CRISPR/Cas9 plasmid
  2. Injection of targeted AB2.2 ES cells into C57BL/6J blastocysts.
  3. Chimera breeding to C57BL/6J
  4. Offspring bred to C57BL/6J
Suggested Control Mice
Wild-type littermates
MMRRC Genetic QC Summary
The MMRRC Centers have developed a genetic QC pipeline using MiniMUGA array genotyping to provide additional information on strain backgrounds for MMRRC congenic and inbred strains. For more information on when data may be available, or to request genotyping for a strain of interest, please contact mmrrc@ucdavis.edu. Older strains may not have this information.
