Strain Development

A vector targeting exon 2 containing LoxP site - FRT site - PGK promoter - Neo cDNA - FRT - distal loxP was transfected into 129S2/SvEvPas-derived ES cells by genOway (France). Two clones were confirmed by PCR and Southern as correctly targeted at the Poln locus, injected into recipient C57BL/6 blastocysts, implanted into pseudo-pregnant females and offspring were assessed for recombinant ES cell contribution by using coat color markers. Two highly chimeric males were generated, mated with C57BL/6 Flp deleter females to allow germline excision of the neomycin selection cassette, which was confirmed by PCR. Progeny were bred a generation further with C57BL/6J mice to generate a pure line of excised floxed alleles, confirmed again by PCR screening and further verified by Southern blot. These mice represent the heterozygous conditional knockout model (B6;129Sv-Polntm1.1Rwd). Males were then mated to C57BL/6 Cre deleter females, offspring were assessed by PCR for excision of exon 2, further verified by Southern blotting. The resulting B6;129S6-Polntm1.2Rwd mouse line was backcrossed 4 times with C57BL/6J, and then random intrastrain-mated heterozygous to heterozygous, and then knockout to knockout for multiple generations.

Note: Recent MUGA analysis revealed 129S6 components.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
