Genetic Alterations

Frame-shift indel mutation on exon 13 of Golgb1 (Gene ID:221439) deleted 41 base pairs. Immunofluorescence staining of E13.5 homozygous embryos confirmed the absence of the Golgb1 protein in these mutants.

Strain Development
We constructed one guide RNA (sgRNA) targeting exon 10 (target sequence 5′-GTCGCCCTCATTATTAACATC-3′) and three sgRNAs targeting exon 13 (target sequences exon13-1: 5′-GCTGCACTTCTCTACGGTCG- 3′, exon13-2: 5′-CACCGCTGGTACTACTCTGCTCAAC-3′, and exon13-3: 5′-CACCGCTGCAGGGCAAGAGGCCCGA-3′) using the online CRISPR Design Tool. The target sequences were subcloned into the PX459 mammalian Cas9-expression vector (Addgene, #48139) and tested by transfection into cultured immortalized mouse embryonic kidney (MK4) cells Subsequently, sgRNAs targeting exon 10 and exon 13, respectively, were synthesized in vitro and co-injected with Cas9 mRNAs into zygotes from FVB/NJ inbred mice to generate genetargeted mice in the CCHMC Transgenic Animal and Genome Editing Facility. Transgenic founder mice were identified by sequencing PCR products and were bred to FVB/NJ inbred mice to test for germline transmission. Since homozygous mice died at birth due to cleft palate, the colonies are maintained through breeding heterozyous males to FVB females (G4, FVB backgroud).

Mice in FVB backgroud seems more aggressive than in other strain backgroud. The grouped mice (especially males) fight each other sometimes after the cage change.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
