Genetic Alterations
Two loxP sites were inserted flanking exon 3 of the CD1d1 gene. Both loxP sites were positioned in the intron upstream and downstream of exon 3. An frt-flanked neomycin selection cassette was removed via FLP mediated recombination.

Strain Development
  • Targeting vector based on vector generated by Gene Bridges Co, (Heidelberg, DE) containing a neo selection cassette flanked by FTR sequences (FRT-pGK-Neo-FRT) and a BAC clone (RP23-181A1) insertion containing LoxP-exon 3-LoxP flanked by ~ 6 kb homologous arms of murine gene Cd1d1 (NCBI Gene ID 12479).
  • Electroporation of linearized target vector into C57BL/6N ES cells (made in-house at Clare Hall and are termed ’B6N6.0’).
  • G418 selection, clonal expansion, and screening via long-range PCR of 100 clones. Three positive clones were identified and further characterized by Southern for correct targeting into the Cd1d1 locus and karyotype analysis. Two out of three clones displayed >90% euploidy. 
  • Two ES clones were injected into blastocysts; implantation into a C57BL/6 female to produce chimeras.
  • Tail snip PCR to confirm genotype of chimeras. TaqMan genotyping assay was developed and performed by Transnetyx (TN).
  • Mating of chimera offspring (high level of chimerism) with C57Bl6 albino (J/N mix) for germline transmission.
  • Mating of mice positive for neomycin cassette with FLPo mice (B6.Cg-Tg(Pgk1-flpo)10Sykr/J - JAX Strain #011065) to remove the neomycin cassette. 
  • Maintenance on C57BL/6J (JAX Strain #000664) background.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
