Genetic Alterations
Exon 28 was replaced with a floxed neo cassette and a modified exon 28 in which nucleotide substitution results in the amino acid substitution of alanine for proline at positions 878 and 881 (P878A, P881A). Cre-mediated recombination removed the neo cassette.

Strain Development
Gene targeting in 129P2/OlaHsd-derived E14Tg2a mouse embryonic stem cells was performed with the use of ESGRO. Chimeric mice were identified by coat color and then bred to C57BL/6J mice. Transmission of the germline was identified by PCR and confirmed by Southern blotting. Heterozygous targeted mice bearing PA[neo] allele (+/PA[neo] mice) were obtained and then crossed with EIIa-Cre mice, which express Cre in the very early stage of embryogenesis, to delete the neomycin cassette to avoid its possible interference with FAK gene expression. The progenies with neomycin cassette removed (+/PA; EIIa-Cre mice - JAX Strain #003724) were crossed with C57BL/6J mice to segregate the FAK PA allele from the heterozygote EIIa-Cre transgene. The resulting heterozygous and homozygous FAK P878A/P881A mutation knock-in mice (+/PA and PA/PA mice) were identified by PCR analysis and confirmed by sequencing of tail DNA.

The line is FVB congenic (N8) with additional components from 129P2 (due to the use of E14TG2a ES cells) and C57BL/6 from some initial breeding (not further specified).
