These mice can be bread into tissue-specific Cre transgenic mice to generate tissue-specific Caspase-8 knock-out mice. In the original paper, the mice were bread into an Lck-Cre mouse which generated a T-cell Caspase-8 null mouse highlighting the essential role of Caspase-8 in T-cell proliferation, immune responses, and in preventing immunodeficiency.

Strain Development
A targeting construct was generated in which exons 3 and 4 of caspase 8, a neomycin resistance cassette and the herpes simplex virus thymidine kinase gene were each flanked by LoxP sites. The linearized construct was electroporated into E14K ES cells, and G418 resistant clones were selected. Clones with an insertion of the targeting vector by homologous recombination at the caspase 8 locus were obtained. To remove the floxed PGKneor-PGKtk cassettes, two independent Casp8fl3-4;neotk/wt ES clones were transiently transfected with a CMV-driven construct encoding Cre recombinase. Clones that had lost the PGKneor-PGKtk cassettes but retained the floxed caspase 8 exons 3 and 4 (Casp8fl3-4) were injected into C57BL/6 blastocysts to generate chimeric mice. Heterozygous (Casp8fl3-4/wt) mutant mice were generated by crossing chimeras to C57BL/6J mice and were subsequently backcrossed into C57BL/6J mice from Jackson (JAX Strain #000664).

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
