Genetic Alterations
The peptide transporter 1 (PepT1), also known as solute carrier family 15 member 1 (SLC15A1), is a protein that, in humans, is encoded by SLC15A1 gene, which mediates the cellular uptake of di-/tripeptides in addition to several peptidomimetic drugs. In this line, DNA sequence bases encoding amino acid residues 16-24 were replaced with an IRES-lacZ-neomycin selection cassette. The transgenic construct stems from a human bacterial artificial chromosome (BAC) RP11-782G13 containing the human SLC15A1 (or PEPT1) gene including 5' regulatory elements, coding area, and 3' regulatory elements. Founder #1 presented with one copy of the human BAC.
Phenotype
The proton-coupled oligopeptide transporter PEPT1 (SLC15A1) is abundantly expressed in the small intestine, and found to mediate the uptake of di/tripeptides and peptide-like drugs from the intestinal lumen. The humanized huPePT1 mouse strain has no obvious behavioral or pathological phenotype. The mRNA and protein profiles indicated that huPEPT1 mice had substantial PEPT1 expression in all regions of the small intestine (i.e., duodenum, jejunum, and ileum) along with low but measurable expression in both, proximal and distal segments of the colon. These findings suggest that huPepT1 mice may provide a valuable tool in the drug discovery process by better predicting the oral pharmacokinetic profiles of PepT1 substrates in humans.

Strain Development
In brief, bacterial artificial chromosomes (BACs) containing the PEPT1 gene (SLC15A1 - NCBI Gene:6564) were obtained from a human BAC library (Empire Genomics, Buffalo, NY). A BAC clone [RP11-782G13; ∼179 kb; CHR13 (98,091,462–98,270,723)], containing the entire 5′-terminal regulatory elements, coding area, and 3′-terminal regulatory elements, was then microinjected into the male pronucleus of fertilized one-cell embryos obtained from Pept1–/– mice on a C57BL/6 background. The pronuclear stage embryos were then transferred into the uterus of pseudopregnant recipient animals. Founder mice were screened to identify an animal containing one copy of the human BAC after which these animals were bred (i.e., mPept1–/–/hPEPT1+/– × mPept1–/–/hPEPT1–/– mice) to maintain hemizygous transgenic lines.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients.
