Genetic Alterations
Using CRISPR/Cas9 gene editing, we targeted the Topologically Associated Domain (TAD) boundary for deletion in order to investigate its functionality as a non-coding regulatory element of the genome. The deletion was approximatively 21,240 bp in size (chr5:120,087,010-120,108,250) and encompasses the boundary of a predicted TAD element. This TAD boundary knockout line is part of the ENCODE project

Phenotype
No overt phenotype observed yet. Adult mice have been sent to the Comparative Pathology Lab at UC Davis for necropsy and histopathologic evaluation.
Strain Development
The Tbx5 TAD boundary knockout mice were generated via CRISPR/Cas9 genome editing by microinjecting into fertilized FVB/NCrl eggs. Founders harboring the correct modification were mated to obtain heterozygous animals for phenotyping. Subsequent generations were crossed in sib-matings for colony expansion and maintenance. Sperm from heterozygous males has been cryopreserved in-house and live animals will be sent for strain archiving into the repository.
Suggested Control Mice
From crossing that generated the experimental animals, littermates that did not harbor the desired mutations were considered "wild-type controls". The background of the control littermates is also FVB/NCrl.

The MMRRC Centers have developed a genetic QC pipeline using MiniMUGA array genotyping to provide additional information on strain backgrounds for MMRRC congenic and inbred strains. 1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
