Genetic Alterations
The transgene is a single-chain version of serine palmitoyltransferase with its three core (human) subunit cDNAs, SPTLC2, SPTSSA, and SPTLC1 fused into a single reading frame linked T2A-separated EGFP under control of a promoter that is activated by Cre recombinase expression (Stop-fSPT). The transgene was inserted into the Gt(ROSA)26Sor locus using TARGATTTM technology (Applied Stem Cell, Milpitas, CA).

Phenotype
Overexpression of the first enzyme of the sphingolipid metabolism pathway. Cre-mediated inducible elevation of sphingolipid synthesis. Per donor, at the time of the archiving, this is the only mouse line that allows inducible expression of the sphingolipid synthesis pathway.
Strain Development
To generate mice to enable the conditional expression of fusion SPT (fSPT), the three individual SPT subunits (in the following order: SPTLC2, SPTSSA, and SPTLC1) were cloned as a single-chain coding unit linked to EGFP by a T2A peptide sequence (Figs. 4 and S1A, PMID:31880535). A stop cassette flanked by LoxP sites was inserted between the CAG promoter and the fSPT coding unit. The expression cassette was inserted into the Rosa26 locus using TARGATT methodology originally developed on FVB. Subsequent backcrossing to C57BL/6J (N7).

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
