    Strain Development
    To generate C9orf72loxP mice, the targeting construct was designed to insert an Frt-flanked neomycin cassette as wells a one loxP site upstream of exon 4 and one loxP site downstream of exon 5. Deletion of exons 4 and 5 targets all reported C9orf72 isoforms in the mouse. This construct was electroporated into C57BL/6J embryonic stem cells. Correctly targeted stem cells (PCR/Southern blot) were injected into blastocysts, and chimeric mice were bred with C57BL/6N mice. The resulting C9orf72loxP-neo mice were then bred with mice expressing Flp recombinase in their germline (B6;SJL-Tg(ACTFLPe)9205Dym/J (RRID:IMSR_JAX:003800) to remove the Frt-flanked neomycin cassette, generating C9orf72loxP/+ offspring. The C9orf72-loxP mice were then backcrossed 10 generations with C57BL/6N, and maintained heterozygous with C57BL/6N.

If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

    2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

    3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
