Genotype Determination
Phenotype

The interleukin 1 beta gene (Il1b) is a potent pro-inflammatory cytokine that is critical for the in vivo survival, expansion, and effector function of IL-17-producing helper T (T(H)17) cells during autoimmune responses. To create the Il1bdel286 allele, CRISPR/Cas9 endonuclease-mediated genome editing was used to generate an exon 3 deletion of Il1b exon 3. In wild-type mice, IL-1b is upregulated in activated macrophages in response to LPS stimulation. IL-1b KO homozygous mice exhibit a dramatic decrease in secretory IL-1b levels as compared to wild-type after 24 hrs and 48 hrs of LPS stimulation. Additional characterization of these Il1bdel286 mice may be added at a later point. Both heterozygous and homozygous mice are viable and fertile.

Strain Development
Guide RNAs were selected to flank exon 3 of the interleukin 1 beta gene. Four guides, Il1b_up_sgRNA1: CAGATTCTGGGCCAGGTAAG, Il1b_up_sgRNA2: AGATTCTGGGCCAGGTAAGT, Il1b_down_sgRNA1: TATGGGACTGCCTTGCTCCC and Il1b_down_sgRNA2: CTCCCAGGCTATTTTGTGAT were used to generate a 286-nucleotide deletion that includes all of exon 3 (CCAGAGCACCCA //del286//AACGGGAAGGGA). Loss of exon 3 is predicted to generate a null allele. These guides and Cas9 nuclease were introduced into single-cell C57BL/6J zygotes and transferred to pseudopregnant females. Progeny were screened by DNA sequencing to identify correctly targeted pups, which were then bred to C57BL/6J mice for germline transmission. The Il1bdel286 colony was backcrossed to C57BL/6J for at least two generations.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
