Genetic Alterations

A targeting vector was designed to insert a VH11DHJH1 gene segment into the locus, with a floxed neo. The neo was subsequently removed via cre-mediated recombination.

Curation note: The data provided for this strain are incomplete and cannot be uniquely tied to genetic entities. The identity of the Ighb allele, for example, is uncertain; it may be a synonym for Ighg2ab (MGI:6515767). Please refer to the cited publications form more information.

    Strain Development
Gene targeting was performed by inserting a Vh11DhJh1 gene segment cloned from a Vh11/Vk9 anti-PtC hydridoma (2-2G4) into a Jh locus knock-in construct that contains the neomycin resistance gene flanked by loxP sites, then transfecting the ES line RW4 (derived from 129X1/SvJ). Chimeric male mice were bred, and upon obtaining germline transmission, Vh11+ mice were crossed with Cre deleter mice to eliminate the neo gene. Vh11+NeoR- offspring were then crosssed to CB17 mice for 8 generations.
    MMRRC Genetic QC Summary
The MMRRC Centers have developed a genetic QC pipeline using MiniMUGA array genotyping to provide additional information on strain backgrounds for MMRRC congenic and inbred strains. For more information on when data may be available, or to request genotyping for a strain of interest, please contact mmrrc@med.unc.edu. Older strains may not have this information.
