Genetic Alterations
This Oprm1-specific Cre mouse line was generated by inserting a T2A cleavable peptide sequence and the Cre coding sequence into the Oprm1 3’UTR using a CRISPR methodology. Importantly, this line shows specificity and fidelity of Oprm1 expression throughout the brain and with respect to function. No differences in behavioral responses to morphine are observed when compared to wild-type mice. No alterations in Oprm1 gene expression or receptor density are detected.

    Strain Development
    A targeting construct encoding a T2A cleavable peptide and Cre recombinase was inserted downstream of exon 4 in the Oprm1 gene sequence. The two guide RNAs were synthesized by in vitro transcription using T7 RNA polymerase. The T7 promoter and sgRNA sequences were amplified by PCR, resulting in a ~ 120 bp template. The sgRNAs were purified using an RNA affinity filter and quantified using capillary electrophoresis. ‘Injection mix’ was prepared containing the two sgRNAs at a final concentration of 50 ng/ml, biotinylated targeting template 20 ng/ml and mRNA encoding the Cas9-mSA nickase at 75 ng/ml. The injection mixture was microinjected into C57BL/6NTac two-cell embryos and cultured in vitro in the presence of 50 nM SCR7. Embryos were placed in this environment for 24 hours to allow development to the morula stage (8 to 16 cell embryo). Embryos were then implanted into the uterus of pseudo pregnant mice and potential founder mice born three weeks later. For the Oprm1-Cre line, one founder line was selected, genome sequencing was performed at 50x coverage to identify any mutations relative to the C57BL/6 reference genome. We found no off-target functional variants in the targeted mice.

The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

    2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

    3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
