Genetic Alterations
This cross is between Mertk-/- and PLPeGFP mice.
Mertktm1Gkm : A neomycin resistance cassette replaced sequences including the last exon, which encodes the carboxy terminal end of the kinase domain and the cytoplasmic tail. RT-PCR analysis demonstrated that a mutant transcript was expressed from this allele that lacked sequences encoding the cytoplasmic kinase domain.

Tg(Plp1-EGFP)10Wmac: A mouse Plp1 promoter was used to drive expression of EGFP.

    Phenotype
    Mertk-/-;PLPeGFP mice breed normally, although occasional small litters may occur (3-5 pups). The absence of Mertk results in blind mice due to retinal degeneration and results in defective efferocytosis of apoptotic cells. Oligodendrocyte precursor cells (OPCs) from neonatal pup brains fluoresce green under PLPeGFP reporter expression and do not express functional Mertk.

    Conditional Phenotype: When subjected to cuprizone-induced demyelination and remyelination, Mertk-/-;PLPeGFP mice exhibit altered phosphoproteome and secretome profiles that impede OPC differentiation, delay myelin debris clearance, and result in significantly slower remyelination compared to wild type mice. Despite this, Mertk-/-OPCs efferocytose apoptotic cells similarly to wild type OPCs. This suggests OPC efferocytosis is mediated by Axl and that Mertk does not play an essential role.

    Strain Development

    B6J.129P2(D2)-Mertktm1Gkm/Mmnc. (RRID:MMRRC_075744-UNC): A 574 bp cDNA mertk fragment was used to screen a 129/Sv genomic library. The tyrosine kinase domain in Mertk cytoplasmic tail was replaced with neomycin resistant gene. 129P2/OlaHsd E14TG2a embryonic cell line was targeted and resulting mice were bred initially to (C57BL/6 x DBA/2 )F1. The targeting resulted in unstable mRNA and lack of the Mertk protein. Mertk null mice were subsequently backcrossed to C57BL/6J for at least 12 generations and maintained by sibling matings.

    B6;CBA-Tg(Plp1-EGFP)10Wmac/J (RRID:IMSR_JAX:033357): The transgenic construct contains the mouse proteolipid protein (myelin) 1 (Plp1) promoter regulatory elements and the Plp1 3' untranslated region ligated to EGFP sequence and injected into C57BL/6 x CBA fertilized oocytes. Founder line 10 was subsequently established. Founder animals were bred to C57BL/6J mice and then backcrossed to the same for at least four generations by the donating lab (see SNP note below). Upon arrival, mice were bred to C57BL/6J inbred mice (Stock No. 000664) for at least one generation to establish the colony.

    Tg(Plp1-EGFP)10Wmac/J was mated to B6J.129P2(D2)-Mertktm1Gkm/Mmnc and homozygous mice were developed. Mertk-/-;PLPeGFP were developed to study myelinating oligodendrocytes in the central nervous system and peripheral Schwann Cells in the absence of Mertk.
