Phenotype

This TG-Gtf2ird1-HA transgenic line overexpress Gtf2ird1. The transgenic insert used includes a human influenza hemagglutinin tag (HA-tag). It may be useful in studies related to Williams Syndrome, a rare neurodevelopmental disorder exhibiting cognitive and behavioral abnormalities, including increased social motivation, risk of anxiety and specific phobias along with perturbed motor function.

Strain Development
A bacterial artificial chromosome which contained the entirety of the 100 kb Gtf2ird1 gene, and 89 kb of flanking regulatory sequence (60 kb at the 5’ end and 28 kb at the 3’), but no additional intact genes or their promoters was utilized to generate the transgene. This was then recombined using standard methods to insert an HA tag in-frame directly before the stop codon of the beta isoforms 208,209. Homologous recombination via transient expression of RecA was employed, followed by Neomycin selection of an inserted FRT-flanked cassette. The selection cassette was then removed after the expression of FLPe, leaving a single FRT site downstream of the stop codon. Transgenic mice overexpressing Gtf2ird1 (TG-Gtf2ird1-HA) were created by injecting this modified BAC into C57BL/6NTac mouse oocytes and transplanting eggs into pseudo pregnant surrogates to carry them to term. Mice were backcrossed to C57BL6/J for over 8 generations prior to commencing experiments.

MMRRC Genetic QC Summary
The MMRRC Centers have developed a genetic QC pipeline using MiniMUGA array genotyping to provide additional information on strain backgrounds for MMRRC congenic and inbred strains. For more information on when data may be available, or to request genotyping for a strain of interest, please contact mmrrc@missouri.edu. Older strains may not have this information.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
