Genetic Alterations
The human STMN2T transgenic construct contains the entire human STMN2 gene from bacterial artificial chromosome clone, RP11-761J24 and including STS sequences corresponding to the 5’ and 3’ extents of STMN2. The BAC clone also includes The BAC was microinjected into the pronuclei of fertilized C57BL/6J oocytes. Founder line 9439 carries 2-3 copies of the transgene. The transgene integrated on Chr.7 [109,434,498-109,451,680; build mm10] the insertion results in a 17 kb deletion of genome between the two identified breakpoints. The deleted sequences include Stk33 intron 1, the entire 1700095J03Rik locus and Trim66 exon 21.

Phenotype
Stmn2delEx3 knock-out mice were created using CRISPR/cas9 genome editing to delete exon 3. The deletion results in the absence of protein and a high degree of homozygous perinatal lethality with 18% of homozygotes surviving to 3 weeks (MMRRC Stock 69524). Hemizygous hSTMN2 transgenic mice express human STMN2 at 50% of wild type levels in the brain and 80% of WT levels in spinal cord. Mice hemizygous for the transgene are viable and fertile. The donating investigator does not recommend maintaining the transgene as a homozygote.When the Stmn2delEx3 and hSTMN2 alleles are combined such that mice are homozygous for Stmn2delEx3 and hemizygous for the human STMN2 transgene, the transgene rescues double mutant mice from the lethality phenotype.

This strain is the result of a cross between mice carrying the Stmn2delEx3 allele (MMRRC Stock 69524) and mice carrying Tg(STMN2)9439Lutzy. Both alleles are maintained on the C57BL/6J background.

Maintained as a homozygous (Stmn2^delEx3) hemizygous (Tg) x heterozygous (Stmn2^delEx3) wild type (Tg) and the reciprocoal mating. The donating investigator does not recommend maintaining the transgene as a homozygote. Mice homozygous for Stmn2^delEx3 in the absence of the transgene die by 3 weeks.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
