Strain Development
The targeting plasmids were linearized and transfected into CJ7 (129S1/Sv-Oca2+ Tyr+ Kitl+) embryonic stem cells and selected using a neomycin resistence casette flanked by FLPase recombination sites. Embryonic stem cell clones were injected into C57BL/6 blastocysts to generate chimeric progeny. Male chimeras were backcrossed to C57BL/6 females. Germline transmission in F1 heterozygous offspring was verified by Southern blot analysis. F1 heterozygotes were interbred with FLPase mice to remove the neomycin casette, and backcrossed with C57BL/6J mice for at least 9 generations before making homozygous conditional mice.
