Genetic Alterations
A 1,850 bp fragment of the human mesothelin promoter drives expression of the SV40 large T antigen. This transgene is expressed in mesothelial cells. Transgenic line 304i carries 32 copies of the transgene.

Phenotype
SV40TAg is expressed in all mesothelial cells in MexTAg mice to varying levels depending on the MexTAg line. Mice carrying high copy numbers of SV40 TAg (304Hom; 64 copies and 299Het 97 copies) have moderate to severe phenotype changes. High copy number mice exhibit thin fur and a hunched back. They are also more prone to abscesses, ear infection, spontaneous tumors, limb, neck, or chest swelling. 299Het mice are prone to anal prolapse and skin tags. Heterozygous lines for 266, 270 and 304 display less severe phenotypes. However, these are not actively bred. All lines except 299h can be maintained as homozygotes. Upon asbestos exposure, 83%-100% of MexTAg mice will develop mesothelioma, with few, if any non-mesothelioma tumors. Minimal tumor development is observed in the absence of asbestos.

Strain Development
A 2,148-bp fragment containing the TAg open reading frame (ORF; SV40 strain 776) was generated with BglII and XbaI restriction endonuclease sites by PCR. The fragment was cloned downstream of 1,850 bp of the mesothelin promoter in pGL3MP1850 to generate pMexTAg. A 4.3-kb ClaI fragment, encompassing the promoter and ORF, was inserted into C57BL/6J embryonic stem cells. Founders were identified by Southern blot and bred and maintained as heterozygotes. The 304i carries 32 copies of the transgene.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
