Phenotype
Inducible knockout of the Kif20a gene in neural progenitor cells causes early cell cycle exit and precocious neuronal differentiation and result in ventriculomegaly in the cortex.
Strain Development
Derivation of the Kif20atm1a knockout-first line:
  • Targeting vector prepared by EUCOMM: PG00127_X_2_E02 containing the L1L2BactP promoter Driven cassette based on backbone R3R4_pBR-amp with LoxP sites flanking exons 5-15 of the Kif20a gene. The linearized targeting vector was transfected into JM8.N4 ES cells (origin: C57BL6/N) by EUCOMM [Project 120814].
  • Three karyotyped Kif20a recombinant clones GEPD0581_C07, D05, and H04 were purchased from the EUCOMM repository.
  • All three ES clones were microinjected into albino blastocysts that were implanted into pseudo-pregnant albino females.
  • Coat color chimeras were generated with all three of the mESC clones.
  • Chimeric mice were bred with (B6(Cg)-Tyrc-2J/J albino mice JAX Strain #000058) to test for germline transmission.
  • Chimeras produced with mESC line GEPD0581_ H04 transmitted the Kif20atm1a mutant allele to their offspring, and the presence of the knockout-first cassette within the Kif20a gene was confirmed by long-range PCR.
  • Maintenance on C57BL6/J (JAX Strain #000664) background.
Derivation of the Kif20atm1c conditional ready line:
  • Kif20atm1a knockout-first allele was converted to a conditional-ready allele by breeding the Kif20a KO-first mice to B6.Cg-Tg(ACTFLPe)9205Dym (JAX Strain #005703) mice, and selecting for mice with LoxP sites flanking Kif20a exons 5-15.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
