Genetic Alterations

Angptl4fl mutant mice contain a conditional knockout of the Angpl4 gene. Using a CRISPR-targeting approach, loxP sites were inserted flanking exons 2 and 3.

Phenotype

Angptl4fl mice have a wildtype phenotype. When they are crossed with AdipoCre to generate an adipose-specific knockout, mice manifest lower plasma triglycerides and higher adipose lipoprotein lipase activity. When crossed with Albumin-Cre mice to generate a liver-specific knockout mice, plasma triglyceride levels are normal unless mice are fasted overnight or longer. With a longer fast mice manifest lower plasma triglyceride levels.

Strain Development
CRISPR guides, Cas9 protein, and a homology donor long single stranded DNA were microinjected into B6SJLF1/J zygotes and progeny were screened for the appropriate insertion of 5′ and 3′ LoxP sites by PCR amplification of the genomic region and subsequent sequencing. A male mouse containing both 5’ and 3’ LoxP sites was mated to C57BL/6J breeders (Jackson Labs Stock #000664) to produce male and female heterozygote mice, identified by PCR. Heterozygous progeny were interbred to generate homozygous floxed mice for initial phenotyping experiments. For cryopreservation and ongoing studies, heterozygote mice were backcrossed to C57BL/6J mice (Jackson Labs Stock #000664) for 10 generations. Then heterozygote mice were interbred to generate homozygous floxed mice.
Suggested Control Mice
For most experiments homozygous floxed mice without Cre are used as controls for homozygous flox Cre positive mice. To examine phenotypes of the conditional allele before Cre recombination, wild-type mice from a heterozygous flox mouse cross would be the control.

The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
