Genetic Alterations
Poglut3 targeting vector was purchased from EuMMCR. Poglut3 tm1a heterozygous ES cells were generated using ES cell line: Nagy G4 (129/SvEvBrd x C57BL/6); JM8A3 (C57BL/6N). Animals heterozygous for the Poglut3 tm1A allele were generated by injecting ES cells into C57BL/6N blastocysts, and resultant chimeras were mated to females of the same strain of mice. Poglut3 tm1.3 (C) alleles were generated using the mouse codon optimized FLP (FLPo) flippase (B6.129S4-Gt(ROSA)26Sor tm2(FLP*)Sor /J.

This strain is the Poglut3 tm1.2 for the global floxed; reporter and neo removed allele.

    Strain Development
    The PG00252_Z_1_B04 targeting vector Poglut3tm380258 was inserted upstream of exon 2 in the Poglut3 locus on chromosome 9. The cassette is composed of an FRT site followed by a splice acceptor, IRES, lacZ, polyA site, a loxP site, a neomycin resistance gene, an SV40 polyA, a second FRT site and a second loxP site. A third loxP site is inserted downstream of exon 3. Dr. Robert Haltiwanger (University of Georgia) obtained the targeting vector from EuMMCR (IKMC Project 101611) and had it introduced into JM8A3 embryonic stem (ES) cells and checked for correct targeting at Texas A&M Institute for Genomic Medicine. ES cells were injected into blastocysts, the resulting chimeric males were bred to C57BL/6N females, and Poglut3 tm1Rsh (knockout first) progeny were crossed to B6.129S4-Gt(ROSA)26Sortm2(FLP*)Sor/J (JAX Stock No. 012930) to remove the lacZ reporter and neomycin cassette at the Mouse Transgenic and Gene Targeting Core at Emory University.
