Genetic Alterations
The arms of the Mrgprd targeting constructs were subcloned from a 129/SvJ genomic DNA BAC clone (Genome Systems). The gene deletion constructs eliminated amino acids 20 to 315 from the 321 amino acid coding region of MRGPRD (GenBank accession number AAK91800). A genomic fragment of PLAP was ligated in-frame to a preexisting NcoI site located 19 amino acids downstream from the start codon. To create the gene conserving construct, IRES-EGFPf-rtTA2(S)M2-ACN was ligated into an AscI restriction site engineered immediately 3' of the Mrgprd stop codon. All constructs contained the EMCV IRES followed by rtTA2(S)M2 and a self-excising loxP-flanked pol-II promoter-neomycin resistance cassette (ACN). Homologous recombination was performed in mouse CJ7 embryonic stem (ES) cells following standard procedures. Correctly targeted ES cell clones were identifed by Southern blot hybridization using probes that flanked the 5' and 3' arms of the targeting constructs, as well as an internal neomycin probe.

    Homozygous: A population of small-diameter nociceptive neurons, those expressing Mas-related G protein coupled receptor D, have been made to express human placental alkaline phosphatase, which allows for visualization of all Mrgprd-expressing neurons.

    Strain Development

    A mouse in which the open reading frame of Mrgprd was replaced with a fusion of PLAP was backcrossed to C57BL/6 for 10 generations and crossed to produce homozygous mice.

    A sample from this MMRRC strain was analyzed using the Mouse Universal Genotyping Array (MUGA) and MMRRC computational tools were used to assess the genetic background. A summary of the data can be found here.

The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary.     2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

    3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
