    Strain Development

    A targeting construct was prepared and exon 1 of the mouse Sohlh2 gene was replaced with a neomycin-resistance gene flanked by 5.0 kb (50) and 5.0 kb (30) of homologous DNA sequences. The vector was linearized and electroporated into embryonic stem cells (AB2.1). The embryonic stem cells, identified as heterozygous for the targeted Sohlh2 mutation, were microinjected into the C57BL/6 blastocysts to produce chimeric mice that carried the mutation into the germline. These mice were mated with C57BL/6 wild-type (WT) mice to generate Sohlh2 heterozygous animals that were subsequently crossed to produce offspring from the second generation for analysis.

    A sample from this MMRRC strain was analyzed using the Mouse Universal Genotyping Array (MUGA) and MMRRC computational tools were used to assess the genetic background. A summary of the data can be found here.

The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary.
