Genetic Alterations
These CRE3 transgenic mice express Cre recombinase under the control of the rat myosin light chain II ventricular (Mylpf or MLC2v) promoter beginning at E11.5 in the brain. This strain may be useful for generating conditional deletions in neurons and their progenitors.
Phenotype
These CRE3 transgenic mice express Cre recombinase under the control of the rat myosin light chain II ventricular (Mylpf or MLC2v) promoter. Cre activity is observed beginning at E11.5 in the brain and only in cells that are committed to be neurons. Expression is found in the grey matter of the brain, cerebellum, spinal cord, retina, and dorsal and sympathetic ganglia. No expression is observed in the heart. CRE3 mice may be useful for generating conditional deletions in neurons and their progenitors.

Strain Development
The transgenic construct contains Cre recombinase under the direction of a 250 bp fragment of the rat myosin light chain II ventricular (Mylpf or MLC2v) promoter. This transgene was microinjected into fertilized oocytes from a FVB/N female. Line CRE3 was established and crossed to a "dh4" transgenic reporter strain on an unknown genetic background. Cre recombinase activity is observed in brain and eyes. Mice then were crossed to B6.129-Ctnnb1tm2Kem/J. Progeny carrying both alleles were backcrossed to C57BL/6 for more than 10 generations and then bred to homozygosity (see SNP note below). At some point, the strain was crossed to B6.Cg-Capns1tm1Kraj. All other alleles were bred out of the strain. Upon arrival at The Jackson Laboratory MMRRC, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes.


A 32 SNP (single nucleotide polymorphism) panel analysis, with 27 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the rederived living colony at The Jackson Laboratory Repository. One of the 27 markers throughout the genome is segregating, and 3 of 5 markers that determine C57BL/6J from C57BL/6N were found to be segregating. These data suggest the mice sent to The Jackson Laboratory MMRRC were on a mixed C57BL/6J ; C57BL/6N genetic background.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
