Genetic Alterations
This C57BL/6 strain carries a transgene expressing the full-length CD45 protein with a mutation that truncates the cytosolic domain to 12 post transmembrane amino acids. The mutation kills all CD45 tyrosine phosphatase activity in the protein produced from the transgene. The transgene, when transferred to the CD45 knockout background, will only produce CD45 lacking the functions of the cytosolic domain. The details of the transgene mutation are as follows:

Genotype Determination
Phenotype
CD45-minigene transgenic strain ΔC (DeltaC or "dC") is supplied on the C57BL/6 background and thus expresses both the transgene-encoded CD45 and the endogenous C57BL/6 CD45. When the ΔC transgene is transferred to the CD45 knockout C57BL/6 background (JAX Strain #002725), the mice express only a truncated CD45 protein with a deleted cytosolic domain and therefore no functions conveyed by the CD45 cytosolic domain, including protein tyrosine phosphatase activity. Strain ΔC on the CD 45 knockout background has the same severe immunodeficiency phenotype as the CD45 knockout strain, including T and B cell functional and maturational abnormalities. The ΔC strain on the knockout background also has the below average breeding characteristics of the CD45 knockout strain. Normal hematopoietic lineage and maturation-specific isoform regulation of the transgene product is maintained due to the inclusion of the natural introns between exons 3 and 9 in the transgene DNA sequence. The transgene encodes the CD45.1 allelic sequence which is detected by anti-CD45.1 antibodies.

CD45 minigene strain ΔC (Delta C) is one of a series of transgenic strains prepared with the CD45 minigene (MMRRC:44051, 46054, 46256, 46257, and 46258). The minigene was altered to delete the cytosolic domain with only the 12 amino acids immediately carboxyl to the transmembrane domain remaining. The resulting truncated CD45 protein is protein tyrosine phosphatase inactive. The ΔC strain has proven useful for studies of CD45 extracellular domain functions and molecular interactions with regard to lymphocyte activation and maturation. This strain should be also suitable for similar activation and maturation studies of other hematopoietic lineages which express CD45.

Note: The “ΔC” transgene locus is homozygous and on the C57BL/6 background. Thus, the rest of the entire genome, including the rest of the chromosome in which the transgene integrated, is C57BL/6. This means these mice express the transgene-encoded CD45.1 (PTPRCa) as well as the endogenous CD45.2 (PTPRCb) of C57BL/6. For most experimental purposes the “ΔC” transgene locus will need to be transferred to the CD45-null C57BL/6 background. This strain is not provided on the CD45-null background since these mice are poor breeders due to being immunodeficient. The strain is routinely maintained on the wild-type C57BL/6 background and bred onto the CD45 knockout background for experimental purposes.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
