Genetic Alterations
Conditional allele flanks first exon of Utf1 with loxP sites. Cre recombinase-mediated deletion produces a null allele lacking potential promoter region and the coding sequence for 188 aminoterminal amino acids, including the start codon.

Phenotype
Conditional deletion in embryonic germ cells results in reduced gonocyte population in newborn males, defects in spermatogenesis, and reduced sperm counts in young adult mice.

The above-described phenotype is conditional upon recombination with a germline restricted Cre transgene. The floxed allele has no phenotype.
Strain Development
  • Targeting vector based on pBluescript generated by high-fidelity PCR amplification of DNA fragments from a BAC clone (RP23-182C11) template. A LoxP-PGK-Neo-LoxP cassette was subcloned 5' of the Utf1 gene, and an additional loxP sequence was added between exons 1 and 2
  • Electroporation of v6.5 mouse ES cells (derived from B6129S4F1) with KpnI-linearized targeting construct
  • ES cells cultured with G418 selection. 18 of 96 G418-resistant clones screened homologously targeted both arms
  • Two ES clones injected into blastocysts; transferred to pseudopregnant female mice
  • Tail-snip PCR confirmed genotype of chimeras
  • Chimeric males mated with C57BL/6J females produced offspring showing germline transmission of Utf1fl allele
  • Subsequent SNP analyses revealed unknown components; genetic background hence deemed STOCK

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
