Genetic Alterations
Chromosomal inversion on Chr14 during transgene integration leading to loss of Bmp4 expression in embryonic eyes. However, the inversion has not altered any sequences within the transcribed region of the gene. The donor assumes that the inversion disrupted (or positionally separated the promoter from) an enhancer that is required for Bmp4 transcription in the embryonic optic vesicle. In situ hybridization data (available on request - 65582_Data2.pdf) show loss of detectable Bmp4 transcripts in the optic vesicle of a homozygous mutant (-/-) embryo, compared to a +/- heterozygote control.

Anophthalmia, dark grey pigmentation. Reduced fertility, particularly for homozygote/homozygote matings. Homozygotes are slightly runted and have a darker coat color than heterozygotes.

Strain Development
FVB/N embryos were microinjected with a construct containing a tyrosinase minigene inserted into a modified version of the Sleeping Beauty transposon.
